Die Bibliothek entstand Anfang der siebziger Jahre des 17. Jahrhunderts planmäßig auf Betreiben von Herzog Moritz von Sachsen-Zeitz. Die neue, im Bereich der Moritzburg in Zeitz aufgestellte Sammlung, die jedoch nicht mit der herzoglichen Privatbibliothek zu verwechseln ist, setzt sich aus den Beständen der älteren historischen Stiftsbibliothek, die der Herzog dem Naumburger Domkapitel entfremdete, und der auf herzogliche Weisung angekauften Teile der Privatbibliothek des Thomas Reinesius zusammmen. Ausgestattet mit Kapitalien, wurde die Sammlung im 17. und 18. Jahrhundert stark vermehrt. 1718 setzte sich das Naumburger Domkapitel wieder in Besitz der Sammlung.
